Study evaluates kidney stone treatment
We explored how a specific type of treatment called prolyl hydroxylase domain inhibitors, particularly roxadustat, impacts the formation of kidney stones. The study used both lab-based experiments and live mouse models to understand how this treatment might help reduce the damaging inflammation associated with the formation of calcium oxalate crystals in the kidneys.

In the lab, we subjected murine renal tubular cells to roxadustat while exposing them to calcium oxalate crystals. Our findings showed that roxadustat effectively limited the adhesion of these harmful crystals to the cells and decreased levels of various proinflammatory proteins that can worsen kidney damage. Additionally, we looked at the interaction between these cells and immune macrophages, noting that roxadustat reduced the inflammatory response in this group as well.

In our in vivo trials with mice, we injected them with roxadustat and found that those treated with the drug had significantly fewer calcium oxalate deposits in their kidneys compared to those receiving saline. Overall, roxadustat appeared to reduce both kidney inflammation and the formation of stones, suggesting it could be a promising treatment option for preventing kidney stones in humans as well.
